I’ve been doing research in Institute of Biochemistry and Cell Biology,
China Academy of Sciences (SIBCB, CAS) since June this year. During this summer, I’ve been working in Professor XXX’s lab, which researches on how Wnt regulates mammary gland’s development and mammary gland’s tumor genesis. My job here is to first make knockdown and overexpression constructs of some genes we’re interested in to do some in vitro experiments, and then to do some further in vivo tests. I’m happy to find that some of the genes have proved to be related with the pathway we are researching on. At the same time, I’ve been learning some basic and featured techniques of the lab from senior graduates, which is interesting and meaningful.
Through doing research and reading related papers, I’ve got a deeper insight into Wnt signal pathway in regulating somatic stem cells and become more interesting in the research on stem cells and some related fields on development. In the future, I’m interested in finding out the key factors in regulating somatic stem cells’ development and making the result into further
applications in both tumor therapy and medical regeneration.
In terms of your newly published Cell paper, I’ve read it carefully and if you don’t mind, I have some questions to talk about with you as follows:
1. In this paper, you focus on McSC and EpSC to explore. I wonder how to exclude other types of cells for there may be another cell type also secreting key factors to make a triple‐coordination.
2. In Figure 2, you show that after TAM induction, differentiated bulb melanocytes from control and mutant mice show similar expression of melanocyte differentiation markers. According to the previous result, why mutant bulb doesn’t show a higher expression level? Is it because the bulb has an “expression peak” or the induction doesn’t work on differentiated bulb?
3. In Figure 4, you show that Wnt10b has an activation function. But why do you use Wnt7a instead in the further experiments? Is it because Wnt10b cannot be purified easily?
4. Also in Figure 4, panel L, you show an in‐vitro experiment, but why not use in vivo instead for it may be more convincing?

I had research project experience in biostatistics, protein detection, and finally find my destiny in stem cells and development. I have been reading certain research papers in this field and a lot of them are in my schedule. To pursue my PhD research in a top university in US has been my dream and your research area matches my interest, and I am also hoping to learn more in this area.
